You can block it off.
Many manifolds are.
Mine was. No detrimental effect.

If you take a green pen and draw a line on your pic from where red and green lines cross to middle of manifold you have the picture you are looking for

You already have water flow from the water pump to the thermostat housing.
The flow into the inlet manifold of a standard manifold is to warm the inlet air fuel mix. Not needed on a twin 40/45 manifold.

As mentiond. If you want to, you can t into the pipe coming from the heater.
Prsonally. I'd leave it blanked off.

That's the T piece that's apart of the hose.
You need a seperate Y piece in that same green hose that goes up to the heater to create the take
Off for the inlet and still
Go to the heater
